比较了常用的三种提取天花粉蛋白(Tichosanthin,TCS)方法的提取效果,通过SDS-PAGE分析了大苞栝楼籽的蛋白组成和分子质量分布,以及大苞括楼的根、叶、茎和愈伤组织中蛋白组成的异同,同时分析了各个器官中26ku蛋白的氨基酸组成。结果表明,Wong的方法较好;栝楼和大苞栝楼的图谱符合程度为56.3%,图谱特征系数为43.7%;大苞栝楼的各个器官的蛋白组成差异较大,其中多数为低分子质量蛋白,具有很多共同的谱带,而且,根、叶、茎和种子中都具有与TCS和核糖体失活蛋白TCK电泳行为相似的蛋白,氨基酸分析也证明了它们组成上的一致性。从而证明大苞栝楼具有潜在的药用价值。
The extraction effects of three commonly used methods for extracting trichosanthin (TCS) were compared. The protein composition and molecular mass distribution of D. grandis were analyzed by SDS-PAGE, and the roots, leaves, and stems of Da Lou Lou were analyzed. The similarities and differences of protein composition in the callus, and the amino acid composition of the 26ku protein in various organs were analyzed at the same time. The results show that Wong’s method is better; the spectrum conformance degree of Diaolou and Dayulou is 56.3%, and the spectral trait coefficient is 43.7%; the protein composition of each organ of Dayulou is quite different, and most of them are low molecules. The quality protein has many common bands, and the roots, leaves, stems and seeds all have proteins similar to the TCS and ribosome inactivating protein TCK electrophoresis. Amino acid analysis also confirmed their compositional consistency. This proves that Da Yu Lou has potential medical value.
