目的探讨左乙拉西坦(LEV)对癫痫大鼠海马组织中5-羟色胺2A(5-HT2A)受体动态表达及对认知功能的影响。方法 108只Wistar大鼠随机分为对照组(NS)24只、正常给药组(LEV)24只、癫痫模型组(Li-PILO)30只、癫痫给药组(Li-PILO+LEV)30只,分别给予LEV组和Li-PILD+LEV组大鼠LEV 200 mg/kg 1次/d灌胃,连续28 d。采用RT-PCR方法分别在第7、14、28天动态检测大鼠海马组织中5-HT2A受体的表达,Morris水迷宫实验测试大鼠的空间学习记忆。结果 LEV与NS组5-HT2A受体表达水平比较差异无统计学意义(P>0.05);与NS组比较,LiPILO组在第7天时表达水平降低,且随时间延长降低水平越明显,即在第14、28天5-HT2A受体表达水平明显降低(P<0.05);与Li-PILO型组比较,Li-PILO+LEV组5-HT2A受体在第7、14、28天表达水平明显升高(P<0.05)。与Li-PILO组比较,Li-PILO+LEV组的逃避潜伏期明显缩短、穿越平台次数显著增加(均P<0.05)。结论 LEV的抗癫痫作用可能与通过上调5HT-2A受体的表达有关,其在抗癫痫的同时还可以改善大鼠的空间学习记忆。
Objective To investigate the effects of levetiracetam (LEV) on the dynamic expression of 5-hydroxytryptamine 2A (5-HT2A) receptor and its cognitive function in hippocampus of epileptic rats. Methods Totally 108 Wistar rats were randomly divided into control group (NS) 24, normal control group (LEV) 24, epilepsy model group (Li-PILO) 30 and epilepsy group (Li-PILO + LEV) Only, were given LEV group and Li-PILD + LEV group rats LEV 200 mg / kg 1 / d gavage for 28 days. The expression of 5-HT2A receptor in rat hippocampus was detected by RT-PCR on days 7, 14 and 28, respectively. Morris water maze test was used to test the spatial learning and memory in rats. Results There was no significant difference in the expression level of 5-HT2A receptor between LEV and NS (P> 0.05). Compared with NS group, the expression level of LiPILO group decreased on the 7th day and decreased with time The expression level of 5-HT2A receptor in Li-PILO + LEV group was significantly lower than that in Li-PILO group on the 14th and 28th days (P <0.05) Increased (P <0.05). Compared with Li-PILO group, the escape latency of Li-PILO + LEV group was significantly shortened and the number of crossing platform significantly increased (all P <0.05). Conclusion The antiepileptic effect of LEV may be related to the up-regulation of 5HT-2A receptor expression, which can improve spatial learning and memory of rats in addition to antiepileptic.
