目的:观察脂质体对日本脑炎病毒E糖蛋白的佐剂作用;方法:利用改进的振荡法,使亲和层析提取的日本脑炎病毒E糖蛋白包裹于脂质体中,制备出脂质体-E蛋白结合物,并分组免疫BALB/c小鼠;通过ELISA,血凝抑制试验(HI)和中和试验(NT)等指标观察.结果:脂质-E蛋白免疫组的特异性抗体水平明显高于游离E蛋白组;在两批病毒攻击保护性试验中,前者保护率分别为50%和57%,后者分别为38%和30%.结论:脂质体对日本脑炎病毒E糖蛋白有明显的佐剂作用.
OBJECTIVE: To observe the adjuvant effect of liposomes on Japanese encephalitis virus E glycoprotein.METHODS: The eukaryotic expression vector of Japanese encephalitis virus E glycoprotein extracted by affinity chromatography was encapsulated in liposomes by the improved oscillating method to prepare The liposome-E protein conjugates were immunized with BALB / c mice in groups; HI, HI and neutralization (NT) were observed by ELISA. Results: The specific antibody level of the lipid-E protein immunized group was significantly higher than that of the free E protein group. In the two batches of virus challenge protective test, the former protection rates were 50% and 57%, while the latter were 38% and 30%. Conclusion: Liposomes have obvious adjuvant effect on Japanese encephalitis virus E glycoprotein.
