通过同源克隆的方法在TSWV高感烟草品种NC89、TSWV耐病烟草品种中烟100中分别克隆出一个疑似抗番茄斑萎病毒(Tomato spotted wilt virus,TSWV)的基因,分别命名为NtSw-5~(NC89)和NtSw-5~(zy100)。序列分析表明,NtSw-5~(NC89)长3819bp,包含1个完整的读码框,编码1273个氨基酸,理论等电点和相对分子质量分别为5.64和147529.5Da;NtSw-5~(zy100)长3165bp,包含1个完整的读码框,编码1054个氨基酸,理论等电点和相对分子质量分别为6.75和122032.9Da;NtSw-5~(NC89)和NtSw-5~(zy100)均不含信号肽,均无明显跨膜区,二者均含有大多数植物抗性蛋白所共有的CC-(NB-ARC)-LRR典型结构。系统进化树分析表明,NtSw-5~(NC89)与马铃薯假定晚疫病抗性蛋白同源R1A-3近源关系最近,NtSw-5~(zy100)与烟草假定晚疫病抗性蛋白同源R1A-3同型X2近源关系最近。该结果可为烟草番茄斑萎病毒抗性研究和烟草番茄斑萎病毒的防治提供理论基础。
Through homologous cloning, a gene suspected of being resistant to Tomato spotted wilt virus (TSWV) was cloned in tobacco TSWV NC89 and TSWV resistant tobacco varieties, respectively, and named as NtSw-5 ~ (NC89) and NtSw-5 ~ (zy100). Sequence analysis showed that NtSw-5 ~ (NC89) was 3819 bp in length and contained a complete reading frame encoding 1273 amino acids. The theoretical isoelectric point and relative molecular mass were 5.64 and 147529.5 Da, respectively. NtSw-5 ~ (zy100) Length of 3165bp, containing a complete reading frame encoding 1054 amino acids, the theoretical isoelectric point and relative molecular mass of 6.75 and 122032.9Da respectively; NtSw-5 ~ (NC89) and NtSw-5 ~ (zy100) There were no obvious transmembrane domains in the signal peptide, both of which contained the CC- (NB-ARC) -LRR typical structure shared by most plant resistance proteins. Phylogenetic tree analysis showed that NtSw-5 ~ (NC89) had closest genetic relationship with putative late blight resistance protein R1A-3 in potato. NtSw-5 ~ (zy100) had the closest relationship with putative late blight resistance protein R1A- 3 homology X2 near source relationship recently. The results provide a theoretical basis for the research on the resistance of tobacco tomato spotted wilt virus and the control of tobacco tomato spot wilt virus.
