本研究探讨环氧化酶-2(COX-2)在白血病骨髓血管新生中的作用。取初发白血病患者(51例)和正常对照组(18例)骨髓液及骨髓活检组织,采用免疫组织化学方法测定骨髓微血管密度(MVD),ELISA法测定骨髓血清血管内皮生长因子(VEGF)含量,流式细胞术(FCM)检测骨髓细胞内COX-2的表达。结果表明:白血病患者骨髓MVD、VEGF含量、骨髓细胞COX-2阳性率与对照组相比均明显增高,p<0.05。MVD、VEGF、COX-2阳性率三者相关系数分别为0.614,0.423,0.577(p<0.05)。结论:白血病可能同实体瘤一样也是一种血管新生依赖性恶性肿瘤,COX-2可能与VEGF协同促进白血病骨髓血管新生
This study was to investigate the role of cyclooxygenase-2 (COX-2) in leukemic bone marrow angiogenesis. Bone marrow and bone marrow biopsy tissues were collected from 51 patients with primary leukemia and 18 normal controls. MVD was measured by immunohistochemical method. The content of vascular endothelial growth factor (VEGF) The expression of COX-2 in bone marrow cells was detected by flow cytometry (FCM). The results showed that the content of MVD and VEGF in bone marrow and the positive rate of COX-2 in bone marrow cells of leukemia patients were significantly higher than those in control group (p <0.05). The positive rates of MVD, VEGF and COX-2 were 0.614,0.423 and 0.577 respectively (p <0.05). CONCLUSIONS: Leukemia may be an angiogenesis-dependent malignant tumor as well as solid tumors. COX-2 may cooperate with VEGF to promote leukemia bone marrow angiogenesis
