目的:本研究通过冰水灌胃法建立便秘型肠易激综合征(Constipation-predominant irritable bowel syndrome,IBSC)大鼠模型,对IBS-C大鼠采用中药麻枳降浊方进行治疗,观察该方对便秘型肠易激综合征大鼠肠组织5-HT、ICC的影响,探讨其可能的作用机制及作用靶点。方法:将40只雄性Wistar大鼠随机分为5组:正常组、模型组、西药组、中成药组和中药组,药物治疗组分别给予西沙比利、复方芦荟胶囊、麻枳降浊方,模型组给予蒸馏水灌胃,各组大鼠正常饲养,不限食水,持续14天。采用HE染色法观察大鼠肠组织病理学形态,免疫组织化学法观察大鼠肠组织中5-HT表达水平,通过免疫组织化学法观察各组大鼠肠组织中c-kit抗体标记ICC的表达及分布特点。结果:干预性治疗后,与模型组相比,大鼠肠组织病理学形态明显好转,肠道各组织5-HT表达水平显著下降(P<0.05),ICC的IOD及阳性面积有所升高(P<0.05)。结论:麻枳降浊方可能通过改善大鼠肠组织病理学形态、降低肠道各组织中5-HT的表达、提高ICC的IOD及阳性面积,从而改善肠道敏感性及动力异常,达到治疗便秘型肠易激综合征的作用。
Objective: To establish a rat model of constipation-predominant irritable bowel syndrome (IBSC) by means of ice-water gavage and to treat IBS-C rats with traditional Chinese medicine Ma Zhijiang Jiangzhuo Decoction Fang on the constipation-induced irritable bowel syndrome rats 5-HT, ICC, to explore its possible mechanism of action and target. Methods: Forty male Wistar rats were randomly divided into five groups: normal group, model group, western medicine group, Chinese patent medicine group and traditional Chinese medicine group. Drug treatment group were given cisapride, compound aloe vera capsule, The model group given distilled water gavage, normal rats in each group, not limited to water, for 14 days. The histopathological changes of rat intestinal tissue were observed by HE staining. The expression of 5-HT in rat intestinal tissue was observed by immunohistochemical method. The expression of c-kit antibody ICC in intestinal tissue was detected by immunohistochemistry And distribution characteristics. Results: Compared with the model group, the histopathological changes of the intestine in the model group were significantly improved and the expression of 5-HT in the intestinal tissue was significantly decreased (P <0.05), and the IOD and positive area of the ICC increased (P <0.05). Conclusion: Mazhi Jiangzhuo Prescription can improve the intestinal sensitivity and motility abnormalities by improving pathological morphology of intestinal tissue, decreasing the expression of 5-HT in various tissues of the intestine, improving IOD and positive area of ICC, Constipation-induced irritable bowel syndrome.
