稳态时健康人体内甲磺酸帕珠沙星对多索茶碱药动学的影响

目的:探讨稳态时甲磺酸帕珠沙星对健康人体内多索茶碱药动学的影响。方法:采用自身交叉试验设计,8名受试者随机分为2组,单用组多索茶碱给药4d;合用组甲磺酸帕珠沙星和多索茶碱联合用药4d。1周后交换给药。采用HPLC法平行监测合用药前后多索茶碱的血药浓度。结果:两药合用时,多索茶碱的t1/2β明显延长、AUC明显增加、CL/F明显下降。结论:甲磺酸帕珠沙星能抑制多索茶碱在健康人体内的消除,提示两药长期合用时应对多索茶碱进行血药浓度监测。

Objective: To investigate the effect of pazufloxacin mesylate on the pharmacokinetics of doxofloxacin in healthy volunteers. Methods: The self-crossover design was used. Eight subjects were randomized into two groups and treated with doxofylline alone for 4 days. The combination group was treated with pazufloxacin mesylate and doxofylline for 4 days. 1 week after the exchange drug. The plasma concentration of doxofylline was monitored by HPLC before and after the combination therapy. Results: The combination of two drugs, doxofylline t1 / 2β significantly prolonged, AUC increased significantly, CL / F decreased significantly. Conclusion: Pazufloxacin mesylate can inhibit the elimination of doxofylline in healthy volunteers, suggesting that Doxofylline should be monitored for plasma concentration of long-term use of both drugs.
