目的本实验通过温肾补虚方对甲减大鼠的血脂及TSHR表达的影响,探讨中医药治疗甲减引起血脂异常的机制及方法。方法雄性SD大鼠随机分为空白组、造模组,造模组予0.05%丙硫氧嘧啶(PTU)蒸馏水。造模成功后,造模组随机分为模型组、西药组、中低剂量组、中药中剂量组、中药膏剂量组,予相应药物,给药时间4周。免疫组化法检测TSHR的表达;ELISA法检测CHOL、LDL、TSH、T3、T4、FT3、FT4。结果造模后,大鼠血清CHOL、LDL、TSH、TSHR升高(P<0.01),T3、T4、FT3、FT4降低(P<0.01)。治疗后,与模型组相比:中药组、西药组T3、T4、FT3、FT4升高显著(P<0.01);中药组CHOL、LDL显著下降(P<0.05);中药组及西药组TSH极显著下降(P<0.01);中药组TSHR水平显著下降(P<0.01)。结论温肾补虚方能改善甲减大鼠CHOL、LDL、TSHR的含量。中药调节CHOL、LDL的机制可能与降低TSHR的表达水平相关。
ObjectiveTo study the mechanism and method of Chinese medicine treatment of hypothyroidism caused by dyslipidemia by the effect of Wenshen tonic on hypolipidemia and TSHR expression in rats. Methods Male Sprague-Dawley rats were randomly divided into blank group, model group and model group with 0.05% propylthiouracil (PTU) distilled water. After successful modeling, the model group was randomly divided into model group, western medicine group, middle and low dose group, medium dose group, Chinese medicine ointment group, to the corresponding drug, and the administration time was 4 weeks. The expression of TSHR was detected by immunohistochemistry. The levels of CHOL, LDL, TSH, T3, T4, FT3 and FT4 were detected by ELISA. Results After model establishment, serum CHOL, LDL, TSH and TSHR in serum increased (P <0.01), and T3, T4, FT3 and FT4 decreased (P <0.01). Compared with model group, the levels of T3, T4, FT3 and FT4 in Chinese medicine group and western medicine group were significantly increased (P <0.01); CHOL and LDL in Chinese medicine group were significantly decreased (P <0.05) (P <0.01). The level of TSHR in TCM group decreased significantly (P <0.01). Conclusion Wenshen tonic can improve the hypothyroid rats CHOL, LDL, TSHR content. The mechanisms by which Chinese medicine regulates CHOL and LDL may be related to the reduction of TSHR expression.
