[目的]研究RASAL1基因及蛋白在胃癌组织中的表达,并分析其与胃癌临床病理特征的关系,初步探讨RASAL1在胃癌发生发展中的意义。[方法]收集50例胃癌及正常胃组织标本,采用RT-PCR方法检测组织中RASAL1mRNA表达,WesternBlot及免疫组织化学方法检测RASAL1蛋白的表达;并结合胃癌的临床病理资料作相关统计学分析。[结果]①RT-PCR检测显示胃癌组织中RASAL1mRNA表达的平均吸光度值为0.51±0.15,而正常胃组织的平均吸光度值为0.64±0.12,胃癌组织的RASAL1mRNA显著低于正常胃组织(P=0.0004)。②WesternBlot分析表明,胃癌组织RASAL1蛋白表达的平均吸光度值为0.52±0.22,而正常胃组织的平均吸光度值为0.68±0.23,前者显著低于后者(P=0.003)。③免疫组化结果表明,RASAL1蛋白主要在胞浆中表达;50例胃癌组织中RASAL1蛋白的表达分别为12例(-)、23例(+)、13例(++)、2例(+++);而在50例正常胃组织中分别为16例(++)、34例(+++);胃癌组织中RASAL1表达较正常胃组织降低(P<0.05)。④RASAL1蛋白的表达与胃癌患者性别、年龄无显著相关,而与肿瘤大小、分化程度、侵袭深度、淋巴转移及TNM分期相关,随病情进展呈下降趋势。[结论]RASAL1基因在胃癌组织中表达明显下调,且与分化程度及进展阶段相关,提示其可能参与了胃癌的发生发展过程,是胃癌的抑癌基因之一。
[Objective] To investigate the expression of RASAL1 gene and its protein in gastric carcinoma and its relationship with clinicopathological features of gastric carcinoma, and to explore the significance of RASAL1 in the development of gastric carcinoma. [Methods] The specimens of gastric cancer and normal gastric tissue were collected from 50 cases. The expression of RASAL1 mRNA was detected by RT-PCR. The expression of RASAL1 protein was detected by Western Blot and immunohistochemistry. Combined with the clinicopathological data of gastric cancer for statistical analysis. [Results] ①RT-PCR assay showed that the average absorbance value of RASAL1 mRNA in gastric cancer tissue was 0.51 ± 0.15, while the average absorbance value in normal gastric tissue was 0.64 ± 0.12. The expression of RASAL1 mRNA in gastric cancer tissue was significantly lower than that in normal gastric tissue (P = 0.0004) . ②Western Blot analysis showed that the average absorbance value of RASAL1 protein in gastric cancer tissue was 0.52 ± 0.22, while the average value of normal gastric tissue was 0.68 ± 0.23, the former was significantly lower than the latter (P = 0.003). ③ The results of immunohistochemistry showed that RASAL1 protein was mainly expressed in the cytoplasm. The expression of RASAL1 protein in 50 cases of gastric carcinoma tissues was 12 cases (-), 23 cases (+), 13 cases (++), 2 cases (+ ++). In 50 normal gastric tissues, 16 cases (++) and 34 cases (+++) respectively. The expression of RASAL1 in gastric cancer tissues was lower than that in normal gastric tissues (P <0.05). The expression of RASAL1 protein was not related to the gender and age of patients with gastric cancer, but correlated with tumor size, differentiation degree, invasion depth, lymphatic metastasis and TNM staging, with the progression of disease decreasing. [Conclusion] The expression of RASAL1 gene is significantly down-regulated in gastric cancer tissues, and is related to the differentiation stage and progression stage, suggesting that RASAL1 may be involved in the development and progression of gastric cancer and is one of the tumor suppressor genes of gastric cancer.
