目的:观察地塞米松对低温冷冻后视网膜色素上皮(retinalpigmentepithelium,RPE)细胞内Ca2+浓度的影响。方法:在-70℃条件下,冷冻RPE细胞30秒,分别加入40、60、100、150、200μg/ml的地塞米松后,应用Fura-2/AM荧光负荷技术,测定RPE细胞Ca2+浓度。结果:地塞米松在40μg/ml~60μg/ml浓度范围可使冷冻后RPE细胞内Ca2+增高18.6%~29.8%,在150μg/ml~200μg/ml可降低细胞内Ca2+浓度28.4%~35.2%。结论:地塞米松对冷冻后RPE细胞内Ca2+的影响表现为双重作用,即低浓度的促进作用和高浓度的抑制作用,提示临床用药应在冷冻瞬间冲击治疗。
Objective: To observe the effect of dexamethasone on intracellular Ca2 + concentration in retinal pigment epithelial cells (RPE) after cryopreservation. Methods: RPE cells were frozen at -70 ℃ for 30 seconds. After adding 40, 60, 100, 150, 200μg / ml dexamethasone respectively, Fura-2 / AM fluorescence load technique was used to determine the concentration of Ca2 + in RPE cells. Results: Dexamethasone increased the intracellular Ca2 + of frozen RPE cells by 18.6% ~ 29.8% at the concentration range of 40μg / ml ~ 60μg / ml, while the intracellular Ca2 + concentration decreased by 150μg / ml ~ 200μg / ml. 4% ~ 35.2%. Conclusion: The effect of dexamethasone on intracellular Ca2 + in RPE cells after cryopreservation is twofold, that is, the effect of low concentration and the high concentration of dexamethasone can inhibit the RPE cells.
