作者用一种新的Seescan的图象分析仪(128级灰度)测定6例原发性血小板增多症(ET)和3例继发性血小板增多症(RT)患者骨髓中的巨核细胞(MK)的脱氧核糖核酸(DNA)含量。全部患者血小板均>500×10~9/L。除2例在11个月以前接受过降低血小板药物治疗外,其余7例未经过治疗,取右髂后上脊骨髓液0.5ml,放入涂硅的试管内,用EDTA抗凝,制成涂片,空气干燥,甲醇/丙酮固定20分钟。Feulgen染色,保存在暗处,1—2周内作DNA分析。在骨髓涂片尾部,选择MK完全平坦和核的轮廓清晰处进行检测。MK核呈现红-紫色的Feulgen阳性反应。图象中可显示核的平均光密度,用手工描绘核连接的面积,以上二个参数的乘积为核的DNA含量
The authors used a new Seescan Image Analyzer (128-level gray scale) to determine the presence of megakaryocytes (MKs) in the bone marrow of 6 patients with essential thrombocythemia (ET) and 3 patients with secondary thrombocytosis ) Of DNA content.
