目的采用不同方法提取苗药用的土人参根中总黄酮,并比较不同提取方法所得总黄酮的抗氧化活性。方法在单因素试验基础上采用响应面法优化超声波处理后固液比、乙醇体积分数、浸提温度及其交互作用对总黄酮得率的影响,DPPH·、超氧阴离子与羟基自由基清除法考查水浴、超声波辅助和酶辅助提取的苗药土人参根中总黄酮的抗氧化活性。结果苗药用土人参根中总黄酮的最佳工艺为固液比1︰26、乙醇体积分数65%、浸提温度70℃,在此条件下,总黄酮得率为2.620%,与回归方程预测值2.603%符合良好。酶法所得总黄酮的抗氧化活性最强且优于Vc。结论该优化工艺实现了土人参根中黄酮类化合物的高效提取,不同提取方法得到的总黄酮抗氧化活性差异明显。
Objective To extract the total flavonoids of ginseng root using different methods and compare the antioxidant activity of the total flavonoids obtained by different extraction methods. Methods The response surface methodology was used to optimize the effects of solid-liquid ratio, ethanol volume fraction, extraction temperature and their interaction on the yield of total flavonoids by using response surface methodology. DPPH ·, superoxide anion and hydroxyl radical scavenging To investigate the anti-oxidative activity of total flavonoids in the ginseng root of Miao medicinal soil with water bath, ultrasonic wave and enzyme-assisted extraction. Results The optimum conditions of the total flavonoids in the roots of Miao medicinal soil were solid-liquid ratio 1: 26, ethanol volume fraction 65% and extraction temperature 70 ℃. Under this condition, the yield of total flavonoids was 2.620% The forecast of 2.603% is in good agreement. Enzymatic flavonoids obtained the strongest antioxidant activity and better than Vc. Conclusion The optimized process of extraction of flavonoids from Radix Ginseng root is achieved efficiently. The antioxidant activity of flavonoids obtained by different extraction methods is obviously different.
