目的研究大蒜主要活性成分之一的二烯丙基硫醚(diallyl sulfide,DAS)对微囊藻毒素LR(MCLR)诱导的细胞毒性和氧化性DNA损伤的影响。方法体外培养人肝癌细胞株HT17,用1μmol/L MCLR染毒的同时给予DAS(0.02~2 mmol/L)作用24 h,同时设立未处理对照组、单纯MCLR染毒组和单纯DAS处理组。测定乳酸脱氢酶(LDH)漏出率来反映细胞毒性,并应用免疫酶染色法检测8-羟基脱氧鸟嘌呤核苷(8-OHdG)水平。结果单纯MCLR染毒组的LDH漏出率和8-OHdG染色强度明显高于未处理对照组。0.2 mmol/L以上的DAS与MCLR联合处理细胞时,LDH漏出率和8-OHdG染色强度明显低于单纯MCLR染毒组。结论 DAS可能对MCLR诱导的细胞毒性和氧化性DNA损伤具有保护作用。
Objective To investigate the effects of diallyl sulfide (DAS), one of the major active ingredients of garlic, on the cytotoxicity and oxidative DNA damage induced by microcystin LR (MCLR). Methods Human hepatocellular carcinoma cell line HT17 was cultured in vitro. DAS (0.02-2 mmol / L) was administered simultaneously with 1μmol / L MCLR for 24 hours. Meanwhile, untreated control group, MCLR-treated group and DAS-treated group were established. The leakage rate of lactate dehydrogenase (LDH) was measured to reflect the cytotoxicity. The level of 8-OH-deoxyguanosine (8-OHdG) was detected by enzyme-linked immunosorbent assay. Results The leakage rate of LDH and the intensity of 8-OHdG in MCLR-treated group were significantly higher than those in untreated control group. 0.2 mmol / L DAS and MCLR combined treatment of cells, LDH leakage and 8-OHdG staining intensity was significantly lower than the MCLR alone group. Conclusion DAS may have a protective effect on MCLR-induced cytotoxicity and oxidative DNA damage.
