目的 建立一种敏感、高特异性的血吸虫病诊断方法。方法 应用 Cystatin捕获血吸虫吐出物中的半胱氨酸蛋白酶 ,再进行 EL ISA试验检测日本血吸虫 (Sj)病人血清中相应的特异抗体。结果 检测 12 0例慢性血吸虫病人血清 ,84例非疫区正常人血清和 36例华支睾吸虫病人血清 ,Cys-tatin- EL ISA检测 Sj病人阳性率为 98.4 % ,与正常人及华支睾吸虫病人无交叉反应。其敏感性(98.4 % )与 SEA- EL ISA法的敏感性 (93.4 % )比较无明显区别 (P>0 .0 5 ) ,但特异性显著高于后者(P<0 .0 5 )。 Cystatin- EL ISA敏感性和特异性亦均高于 AWA- EL ISA和 ES- EL ISA。结论 Cys-tatin- EL ISA敏感性高 ,特异性强 ,是一种有效的诊断血吸虫病的方法。
Objective To establish a sensitive and specific method for the diagnosis of schistosomiasis. Methods The Cystatin was used to capture the cysteine protease in the exudates of Schistosoma japonicum and the ELISA was performed to detect the corresponding specific antibodies in the serum of patients with Schistosoma japonicum (Sj). Results Serum from 120 patients with chronic schistosomiasis, 84 patients with non-infected normal human serum and 36 patients with Clonorchis sinensis were detected. The positive rate of Cys-tatin-ELISA in detecting Sj was 98.4% Trematodes patients without cross-reaction. The sensitivity (98.4%) was not significantly different from that of SEA-EL ISA (93.4%) (P> 0.05), but the specificity was significantly higher than the latter (P <0.05). The sensitivity and specificity of Cystatin-EL ISA were also higher than those of AWA-EL ISA and ES-EL ISA. Conclusions Cys-tatin-EL ISA is sensitive and specific and is an effective method to diagnose schistosomiasis.
