目的 初步探索神经生长因子低亲和力受体(p75 neurotrophin receptor,p75NTR)在喉癌Hep2细胞系的表达情况并研究其相关生物学功能.方法 应用免疫细胞化学及免疫荧光染色法,检测p75NTR在喉癌Hep2细胞系中的表达;应用RNAi法下调p75NTR表达,通过Real-Time-PCR及Western blot方法验证基因干扰有效,MTT法观察下调p75NTR蛋白水平对喉癌Hep2细胞存活及增殖的影响.结果 喉癌Hep2细胞系内小部分肿瘤细胞显示p75NTR免疫染色强阳性,多数呈强阳性显色的细胞处于旺盛的分裂增殖状态.通过Real-Time-PCR及Western blot方法均证实预先设计的小干扰RNA(siRNA)可以有效下调p75NTR蛋白的表达,转染siRNA抑制p75NTR表达能够抑制喉癌Hep2细胞系的增殖.结论 p75NTR高表达于喉癌Hep2细胞系中具有较强增殖活力的细胞.p75NTR表达对支持和促进喉癌细胞增殖具有重要作用,p75NTR受体信号途径激活与喉癌细胞存活及增殖密切相关,其能否成为肿瘤治疗的靶点有待进一步深入研究.“,”Objective To investigate the expression of nerve growth factor receptor p75NTR in human laryngeal squamous cell carcinoma (LSCC) and its corresponding functions.Methods The expression of p75NTR was tested by immunocytochemistry in human laryngocarcinoma Hep2 cells.Using RNAi method to knock down p75NTR expression,and we confirmed the effectivity by Quantitative Real-time PCR and Western blot test.The influence of p75NTR downregulation on Hep2 cells were studied with MTT method.Results After staining we detected that a small number of Hep-2 cells expressed p75NTR in high level;parts of this strong-positive cells were in multiplicative or division status.By RNAi method we knocked down p75NTR expression,and confirmed effectivity by Quantitative Real-time PCR and Western blot.Down regulation of p75N TR level in Hep2 cells by RNAi could result in proliferation inhibition.Conclusion p75N TR overexpressed in a small group of Hep-2 cells are considered to have high proliferation vitality.Our findings support that p75NTR expression is necessary for survival and proliferation of Hep-2 cells.The activation of p75N TR receptor signaling pathway is closely related to the survival and proliferation of LSCC.We needs further study in whether it can be one target of tumor therapy.
