目的:探讨PELP1与牙周膜细胞骨向分化的相关性。方法:通过对正常人牙周膜细胞与炎症人牙周膜细胞的分离、培养、鉴定,采用RT-PCR、免疫细胞化学、免疫荧光化学及Western blot技术,分别从基因和蛋白水平检测PELP1在两组细胞中的表达及其在细胞增殖和骨向分化过程中的表达变化。结果:在H-hPDLCs和I-hPDLCs中均存在PELP1 mRNA及蛋白的表达。PELP1参与调节hPDLCs的生长与增殖,在H-hPDLCs中与细胞增殖呈正相关,在I-hPDLCs中与细胞增殖呈负相关。PELP1的表达水平在两组hPDLCs骨向分化中亦表现出不同的变化趋势,在H-hPDLCs骨向诱导分化后升高,在I-hPDLCs骨向诱导分化后降低。结论:PELP1在炎症人牙周膜细胞中表达并参与调节细胞的生长增殖及骨向分化。
Objective: To investigate the relationship between PELP1 and bone differentiation of periodontal ligament cells. Methods: The isolation, culture and identification of human periodontal ligament cells and human periodontal ligament cells were studied by RT-PCR, immunocytochemistry, immunofluorescence and Western blot. The expression of PELP1 Expression of two groups of cells and their changes in cell proliferation and differentiation during bone differentiation. Results: The expression of PELP1 mRNA and protein both in H-hPDLCs and I-hPDLCs. PELP1 is involved in the regulation of the growth and proliferation of hPDLCs. It is positively correlated with cell proliferation in H-hPDLCs and negatively correlated with cell proliferation in I-hPDLCs. The expression level of PELP1 also showed different trends in bone differentiation of hPDLCs in both groups. The expression of PELP1 in H-hPDLCs increased after bone differentiation and decreased after bone differentiation. Conclusion: PELP1 is expressed in human periodontal ligament cells and is involved in the regulation of cell proliferation and differentiation.
