目的探讨蓝萼甲素(GLA)对人宫颈癌He La细胞裸鼠皮下移植瘤生长的作用机制。方法建立人宫颈癌裸鼠皮下移植瘤模型并进行随机分组;通过移植瘤生长抑制试验、TUNEL、免疫组化、Western blot和RT-RCR观察移植瘤大体形态、细胞、蛋白及基因表达的变化。结果裸鼠成瘤率为100%;在同一时间内,随着GLA剂量的增加移植瘤的体积逐渐减少,差异有统计学意义(P<0.05);TUNEL法表明随GLA剂量的增加凋亡指数逐渐增多,差异有统计学意义(P<0.05);免疫组化、Western blot检测各组移植瘤中p53及Bax蛋白随GLA剂量的增加表达逐渐增强,差异有统计学意义(P<0.05);RT-RCR法测移植瘤中p53及Bax mRNA的表达随GLA剂量的增加表达逐渐增强,差异有统计学意义(P<0.05)。结论 GLA能明显抑制人宫颈癌He La细胞裸鼠皮下移植瘤的生长,其机制可能与上调P53和Bax的表达有关。
Objective To investigate the mechanism of GLA on the growth of subcutaneous xenografts in nude mice bearing human cervical cancer He La cells. Methods Human cervical cancer xenografts model was established and randomly divided into groups. The morphological, cell, protein and gene expressions of the xenografts were observed by TUNEL, immunohistochemistry, Western blot and RT-RCR. Results The tumor formation rate of nude mice was 100%. At the same time, the volume of xenograft tumor gradually decreased with the increase of GLA dose (P <0.05). The TUNEL method showed that the apoptosis index (P <0.05). Immunohistochemistry and Western blot showed that the expression of p53 and Bax protein increased gradually with the increase of GLA dose in all groups (P <0.05). The expression of p53 and Bax mRNA in the transplanted tumors increased gradually with the increase of GLA dose, the difference was statistically significant (P <0.05). Conclusion GLA can significantly inhibit the growth of human cervical carcinoma HeLa subcutaneously transplanted tumor in nude mice, the mechanism may be related to up-regulation of P53 and Bax expression.
