【目的】探讨激活补体是否会引起心肌缺血损伤 ,以及膜结合型补体调节蛋白CD59对补体引起的心肌缺血损伤有无保护作用。【方法】 18只豚鼠离体工作心脏随机分为 :A组 (对照组 ) ,在灌注液中加入体积分数为 3%灭活人血浆 +酵母多糖 ;B组 ,在灌注液中加入 3%人血浆 +酵母多糖 ;C组 :在灌注液中加入CD59(45 0 μg) +3%人血浆 +酵母多糖。记录各组处理前及处理后 15、30、45和 6 0min的心外膜心电图ST段变化、冠脉流量 (CF)、心输出量 (CO)、左心室最大内压 (LVPmax)、左心室内压最大上升速率 (dp/dtmax)和心率 (HR)。【结果】A组处理前后各项观察指标无明显变化 ;B组在接受处理后心外膜心电图出现ST段抬高、HR增加 ,CF、CO、LVPmax和dp/dtmax均下降 ,这些变化以处理后 45min最明显 ;C组除处理后 45~ 6 0min时心率稍增快外 ,其余指标处理前后无明显变化。【结论】 激活补体可直接引起豚鼠工作心脏心肌缺血损伤 ,CD59对补体介导的心肌缺血损伤有保护作用。
【Objective】 To investigate whether activation of complement may cause myocardial ischemic injury and whether membrane-bound complement regulatory protein CD59 has a protective effect on complement-induced myocardial ischemia. 【Method】 Eighteen guinea pigs were randomly divided into three groups: A group (control group), 3% inactivated human plasma + Zymosan (PBS) in the perfusate, and 3% in group B Plasma + Zymosan; Group C: CD59 (45 0 μg) + 3% human plasma + zymosan were added to the perfusate. The epicardial electrocardiographic ST-segment changes, coronary flow (CF), cardiac output (CO), left ventricular maximum pressure (LVPmax), left ventricular The maximum rate of increase of indoor pressure (dp / dtmax) and heart rate (HR). 【Results】 There were no significant changes in all the indexes before and after treatment in group A; in group B, ST segment elevation, HR increase, CF, CO, LVPmax and dp / dtmax in epicardial electrocardiogram were all decreased 45min after the most obvious; C group except 45 ~ 6min after treatment slightly increased heart rate, the other indicators before and after treatment no significant change. 【Conclusion】 Activation of complement can directly induce myocardial ischemic injury in guinea pigs, and CD59 has a protective effect on complement-mediated myocardial ischemia.
