目的观察多囊卵巢综合征(polycystic ovary syndrome,PCOS)患者外周血中Th17细胞与FOXP3+CD25highCD4+调节性T(Treg)细胞比例及相关因子表达的变化并探讨其意义。方法取8例PCOS患者及12例健康妇女外周血,流式细胞术检测Th17和Treg细胞占CD4+T细胞的比例;实时定量PCR测定外周血单个核细胞(PBMC)中Th17和Treg细胞相关因子IL-17、RORγt、FOXP3、TGF-β、IL-1β及IL-6 mRNA的相对表达量。结果 PCOS患者外周血中Th17细胞的比例明显高于健康妇女(P<0.01),Treg细胞的比例较健康妇女有增高趋势但差异无统计学意义(P>0.05)。PCOS患者PBMCs中,IL-17、RORγt、FOXP3、TGF-β的mRNA相对表达量明显高于健康妇女(P<0.01),而IL-1β则明显低于健康妇女(P<0.01),IL-6差异无统计学意义(P>0.05)。结论 PCOS患者可能处于一种Th17细胞明显增多的炎性状态,这可能参与了PCOS的发生发展,但其中细胞因子的作用尚待进一步研究。
Objective To investigate the changes of the ratio of Th17 cells to FOXP3 + CD25high CD4 + regulatory T (Treg) cells and their related factors in patients with polycystic ovary syndrome (PCOS) and their significance. Methods Peripheral blood was obtained from 8 PCOS patients and 12 healthy women. Flow cytometry was used to detect the ratio of Th17 and Treg cells to CD4 + T cells. The levels of Th17 and Treg cells in peripheral blood mononuclear cells (PBMCs) IL-17, RORγt, FOXP3, TGF-β, IL-1β and IL-6 mRNA. Results The proportion of Th17 cells in peripheral blood of patients with PCOS was significantly higher than that of healthy women (P <0.01). The proportion of Treg cells in healthy women was higher than that in healthy women (P> 0.05). The mRNA expression of IL-17, RORγt, FOXP3 and TGF-βin PCOS patients was significantly higher than that in healthy women (P <0.01), while IL-1β was significantly lower than that in healthy women (P <0.01) 6 difference was not statistically significant (P> 0.05). Conclusions PCOS patients may be in a significantly increased inflammatory state of Th17 cells, which may be involved in the development of PCOS, but the role of cytokines remains to be further studied.
