应用HPLC方法检测人血清中卡铂(Carboplatin)浓度.采用C18分析柱及预柱,流动相为重蒸水,流速1.1ml/min,检测波长为229nm,并用乙醚和氯仿去除内源性杂质干扰。实验结果表明:卡铂与内源性杂质分离良好,保留时间为8.70~9.31min,在血清中卡铂回收率96.49%。卡铂在lμg/ml以上线性关系良好,相关系数r为0.9999,回归方程Y=0.373lX-0.1025,日内及日间变异系数均小于6%(n=5),最低检测浓度为1μg/ml。
HPLC method was used to detect the concentration of Carboplatin in human serum. A C18 analytical column and precolumn were used. The mobile phase consisted of heavy distilled water with a flow rate of 1.1 ml / min and a detection wavelength of 229 nm. Endogenous impurities were removed by ether and chloroform. The experimental results show that carboplatin is well separated from endogenous impurities with a retention time of 8.70-9.31min and a 96.49% recovery of carboplatin in serum. The linearity of carboplatin above 1μg / ml was good, the correlation coefficient r was 0.9999, the regression equation Y was 0.373lX-0.1025, the intra-and inter-day CVs were less than 6% (n = 5) 1 μg / ml.
