目的:优选知母皂苷AⅢ的酶解工艺。方法:采用β-葡萄糖苷酶水解知母皂苷制备知母皂苷AⅢ,以知母皂苷AⅢ转化率为指标,通过单因素和正交试验考察反应温度、反应时间、pH及酶用量对知母皂苷AⅢ酶解工艺的影响。结果:最佳酶解工艺条件为温度55℃,pH 5.0乙酸-乙酸钠缓冲液,酶用量600 U.g-1,反应时间3 h。结论:优选的酶解工艺简单可靠,反应条件温和,适用于工业化生产。
Objective: To optimize the enzymatic hydrolysis process of timosaponin A Ⅲ. Methods: The timosaponin A Ⅲ was prepared by hydrolysis of timosaponins with β-glucosidase. The conversion rate of timosaponin A Ⅲ was used as an index. The reaction temperature, reaction time, pH and enzyme dosage were used to evaluate the effects of timosaponin A Ⅲ A Ⅲ enzymatic hydrolysis process. Results: The optimal enzymatic hydrolysis conditions were temperature 55 ℃, pH 5.0 acetic acid-sodium acetate buffer, enzyme dosage 600 U.g-1, reaction time 3 h. Conclusion: The optimal enzymatic hydrolysis process is simple and reliable, the reaction conditions are mild, suitable for industrial production.
