目的:探讨SLC30A8和CDKAL1基因三个位点rs13266634、rs7756992、rs7754840基因多态性与衢州汉族妇女妊娠期糖尿病(GDM)的相关性。方法:从160例糖尿病妊娠妇女和160例健康孕妇外周血中提取基因组DNA,采用PCR-RFLP法对SLC30A8和CDKAL1基因三个位点rs13266634、rs7756992、rs7754840进行基因分型,明确这些位点与妊娠期糖尿病的关系。结果:SLC30A8基因rs13266634位点KK型和EK型在GDM组的基因频率分别为71.6%和20.0%,高于对照组(59.2%,14.3%),两组相比有统计学意义(P<0.05),E、K基因频率在两组间分布无统计学意义。CDKAL1基因rs7756992位点在两组人群中的基因频率和等位基因频率无统计学差异(P>0.05);rs7754840位点G/A型基因型频率在GDM组为31.7%,对照组为8.2%,两组间有统计学意义(P<0.05),A基因在两组的基因频率分别为16.7%和4.1%,两组间有统计学意义(P<0.05)。结论:SLC30A8基因rs13266634位点和CDKAL1基因rs7754840位点基因多态性可能与GDM发生有相关性。
Objective: To investigate the association of rs13266634, rs7756992, rs7754840 gene polymorphisms with gestational diabetes mellitus (GDM) in Quzhou Han women at three sites of SLC30A8 and CDKAL1. Methods: Genomic DNA was extracted from peripheral blood of 160 pregnant women with diabetes and 160 healthy pregnant women. PCR-RFLP was used to genotype the rs13266634, rs7756992 and rs7754840 sites of SLC30A8 and CDKAL1 genes. Period of diabetes. Results: The gene frequencies of SLC30A8 gene rs13266634 locus KK and EK in GDM group were 71.6% and 20.0%, respectively, which were significantly higher than those in control group (59.2% and 14.3%, P <0.05) ), E, K gene frequency distribution between the two groups was not statistically significant. The frequency of rs7756992 in CDKAL1 gene was not significantly different between the two groups (P> 0.05). The frequency of genotype G / A in rs7754840 was 31.7% in GDM group and 8.2% in control group (P <0.05). The gene frequencies of A gene in the two groups were 16.7% and 4.1%, respectively, with statistical significance between the two groups (P <0.05). Conclusion: The rs13266634 locus of SLC30A8 gene and the rs7754840 locus of CDKAL1 gene may be associated with GDM.
