目的 探讨人脑星形细胞瘤中多种基因蛋白异常表达及意义。方法 应用免疫组化技术检测102例手术切除人脑星形细胞瘤组织标本中增殖细胞核抗原(PCNA)及p53、p21ras及EGFR蛋白的表达。结果 星形细胞瘤组织PCNA标记指数(PC-NALI)为55.6±15.5,p53、P21ras及EGFR蛋白的表达率分别为58.6、52.3%、42%,均显著高于正常脑组织。PCNALI及EGFR蛋白表达与星形细胞瘤恶性程度相关(P<0.01),p53、p21ras蛋白的表达与星形细胞瘤恶性程度无关(P>0.05)。星形细胞瘤中PCNALI与EGFR蛋白表达无明显相关(P>0.05),而与p53、p21ras表达相关(P<0.05)。结论p53、p21rasEGFR异常表达与星形细胞瘤的发生发展相关,RCNALI是评估星形细胞瘤增殖活性良好指标。
Objective To investigate the abnormal expression of multiple gene proteins in human brain astrocytoma and its significance. Methods Immunohistochemistry was used to detect the expression of proliferating cell nuclear antigen (PCNA), p53, p21ras and EGFR protein in 102 cases of resected human astrocytoma. Results The PCNA labeling index (PC-NALI) of astrocytoma was 55.6 ± 15.5, the expression rates of p53, P21ras and EGFR were 58.6, 52.3% and 42% respectively Normal brain tissue. The expression of PCNALI and EGFR was correlated with the malignancy of astrocytoma (P <0.01). The expression of p53 and p21ras was not associated with the malignancy of astrocytoma (P> 0.05). There was no significant correlation between PCNALI and EGFR protein expression in astrocytomas (P> 0.05), and p53, p21ras expression (P <0.05). Conclusions The abnormal expression of p53 and p21rasEGFR is associated with the development of astrocytoma. RCNALI is a good indicator to evaluate the proliferative activity of astrocytomas.
