目的检测反转录病毒介导的HSV-tk/ACV系统的安全性。方法DNA聚合酶链式反应(PCR)、反转录PCR(RT/PCR),Southern杂交及neo基因和LacZ基因补救分析:细胞接种、X-gal染色、TK基因PCR和病理切片检查等。结果从DNA、RNA水平和病毒活体水平对产HSV-tk病毒包装细胞(TK/VPC)、C5转TK基因细胞和实验动物血流中未能检测到具有复制能力的野生型反转录病毒(RCR);PA317/BAG,PA/317/TK产病毒细胞(VPC)于动物体内存活期不超过一个月,未形成肿瘤,无严重炎症反应,HSV-tk/ACV系统及代谢产物无明显毒副作用。结论反转录病毒介导的HSV-tk/ACV系统在动物体内取得了安全的结果。
Objective To test the safety of retrovirus-mediated HSV-tk / ACV system. Methods DNA polymerase chain reaction (PCR), reverse transcription PCR (RT / PCR), Southern hybridization and neo gene and LacZ gene rescue analysis: cell inoculation, X-gal staining, TK gene PCR and biopsy. Results The replication-competent wild-type retrovirus (HSV-tk) was not detected in the bloodstream of HSV-tk virus packaging cells (TK / VPC), C5 transgenic TK cells and experimental animals at the level of DNA and RNA, RCR); PA317 / BAG and PA / 317 / TK virulent cells (VPC) did not survive in the animals for more than one month, did not form tumors, no serious inflammatory reaction, HSV-tk / ACV system and metabolites had no obvious side effects . Conclusion The retrovirus-mediated HSV-tk / ACV system has achieved safe results in animals.
