目的从用于黄鼠性别鉴定的sry和smcY引物中筛选出能够用于喜马拉雅旱獭性别鉴定的引物。方法用1对sry引物和4条smcY引物的3对组合对已知性别的雌雄各10只喜马拉雅旱獭基因组DNA进行扩增,同时,用1对微卫星引物来验证反应体系的有效性。结果在验证了PCR反应有效性的前提下,sry引物从100~1000bp间扩增出多个条带,且未发现雄性特异性条带,smcY引物组合1在雄性旱獭中扩增出100bp左右的亮带,但是部分雌性样品中在600bp左右扩增出较弱的条带,组合2未扩增出任何条带,组合3的所有雄性在250bp处扩增出明亮的条带,雌性个体中未见任何条带。结论 smcY引物组合3可以用于喜马拉雅旱獭性别鉴定。
Objective To screen primers that can be used for gender identification of Himalayan marmot from the sry and smcY primers used for the sex determination of the squirrels. Methods The genomic DNA of 10 Himalayan Marmota male and female with known sex was amplified by 3 pairs of sry primers and 4 smcY primers. At the same time, a pair of microsatellite primers was used to verify the effectiveness of the reaction system. Results After validating the validity of the PCR reaction, sry primers amplified a number of bands from 100 to 1000 bp and no male-specific bands were found. The smcY primer combination 1 amplified about 100 bp in male marmots However, some of the female samples amplified weaker bands around 600bp, while those from combination 2 did not amplify any bands. All the males of combination 3 amplified bright bands at 250bp, and none of the female individuals See any bar. Conclusion smcY primer combination 3 can be used for gender identification of Himalayan marmot.
