目的 研究胰岛素样生长因子 (insulin likegrowthfactorⅠ ,IGF Ⅰ )、碱性成纤维细胞生长因子 (basicfibroblastgrowthfactor,bFGF)、转化生长因子 β1(transforminggrowthfactorβ1,TGF β1)单独及联合应用 ,对人髁突软骨细胞增殖的剂量 效应及时间 效应。方法 采用体外细胞培养技术及噻唑蓝比色法 ,对 3种生长因子对软骨细胞增殖的调节作用进行观察。结果 在培养基中含 0 4%血清条件下 ,TGF β1、IGF Ⅰ的作用不显著 ,bFGF有一定促增殖作用。在含 10 %血清条件下 ,3种生长因子均能显著促进细胞增殖 ,以bFGF最为突出 (增加 6 5 % ) ,生长因子在各自一定浓度范围内呈量效关系。联合应用作用效果累加。生长因子促增殖效应可持续 1周。结论 提示探索最佳生长因子组合可有效促进软骨细胞增殖及基质合成 ,可为关节软骨损伤性疾患的治疗提供一种有效的途径。
Objective To investigate the effects of insulin like growth factor Ⅰ (IGF Ⅰ), basic fibroblast growth factor (bFGF) and transforming growth factor β1 (TGF β1) alone and in combination on the proliferation of human condylar chondrocytes Dose and time effects. Methods The effects of three kinds of growth factors on the proliferation of chondrocytes were observed by in vitro cell culture and thiazolyl blue colorimetry. Results Under the condition of containing 0 4% serum in culture medium, the effect of TGF β1 and IGF Ⅰ was not significant and bFGF had a certain promoting effect. Under the conditions of 10% serum, all the three kinds of growth factors can significantly promote cell proliferation, with bFGF being the most prominent (65% increase). The growth factors were dose-effect relationship within certain concentration range. Combined effect of cumulative effect. Growth factors promote proliferation effect for 1 week. Conclusions It is suggested that exploring the best combination of growth factors can effectively promote chondrocyte proliferation and matrix synthesis, which can provide an effective approach for the treatment of articular cartilage injury.
