低温是影响植物正常生长发育,导致作物产量和品质下降的主要自然灾害之一。由于橡胶树(Hevea brasiliensis)属于热带植物,不耐寒,所以极易受到低温冷害影响。而Inducer of CBF expression 1(ICE1)是低温胁迫信号通路中的重要转录激活因子,目前橡胶树冷胁迫信号途径中包括Hb ICE1在内的大部分关键基因的研究还比较匮乏。我们从橡胶树中克隆了Hb ICE1基因。经测序发现,该基因开放阅读框为1 407 bp,编码469个氨基酸,蛋白质分子质量约为51 k D,理论等电点(p I)为5.83,为亲水性蛋白。并且利用荧光定量PCR技术分析了Hb ICE1在不同激素(乙烯,脱落酸,茉莉酸)以及H2O2处理后的表达模式,结果表明乙烯、脱落酸、茉莉酸以及H2O2均能调控橡胶树Hb ICE1基因的表达,但不同胁迫处理下的表达模式存在着较大的差异,暗示着Hb ICE1在不同的激素信号途径中发挥的作用差异较大。
Low temperature is one of the major natural disasters that affect the normal growth and development of plants and lead to the decline of crop yield and quality. As Hevea brasiliensis belongs to the tropical plants, not cold, so vulnerable to cold damage. Inducer of CBF expression 1 (ICE1) is an important transcriptional activator in low-temperature stress signaling pathways. At present, most of the key genes including Hb ICE1 in cold stress signal pathways of rubber trees are still scarce. We cloned the Hb ICE1 gene from the rubber tree. After sequencing, the open reading frame of this gene was 1 407 bp, encoding 469 amino acids. The molecular mass of the gene was 51 kD and the theoretical isoelectric point (p I) was 5.83, which was a hydrophilic protein. The expression pattern of Hb ICE1 in different hormones (ethylene, abscisic acid, jasmonic acid) and H2O2 was analyzed by fluorescence quantitative PCR. The results showed that ethylene, abscisic acid, jasmonic acid and H2O2 could regulate the expression of ICE1 gene in rubber tree , But there was a big difference in expression patterns under different stress treatments, suggesting that Hb ICE1 may play a different role in different hormone signaling pathways.
