目的:探讨性激素对子宫肌瘤的发生和生长的调节作用。方法:采用体外子宫内膜间质细胞培养,加以雌二醇(E2)、孕酮(P)、睾酮(T)刺激后,进行流式细胞学测定其细胞周期和凋亡的动力学变化。结果:T单独作用时其促进DNA合成及有丝分裂效果最明显,但与E2或P合用时其作用大大削弱;而E2+P对细胞DNA合成及有丝分裂的促进作用比单用E2强,两者的这种协同作用在有T存在的情况下也被减弱。P对细胞的G2+M期作用最强。T对细胞凋亡具有促进作用,T和P之间存在拮抗作用,P能够抑制细胞凋亡。结论:雌孕激素与雄激素之间的代谢平衡对于维持细胞的正常功能至关重要,若局部组织细胞增生与凋亡失衡则会导致肿瘤的发生。
Objective: To investigate the regulatory effect of sex hormones on the occurrence and growth of uterine fibroids. Methods: Endometrial stromal cells were cultured in vitro and stimulated with estradiol (E2), progesterone (P) and testosterone (T). The changes of cell cycle and apoptosis were measured by flow cytometry. Results: The effect of T alone on DNA synthesis and mitosis was most obvious, but its effect was greatly weakened when combined with E2 or P. The effect of E2 + P on DNA synthesis and mitosis was stronger than that of E2 alone This synergy is also reduced in the presence of T. P has the strongest effect on the G2 + M phase of the cells. T has a promoting effect on apoptosis, antagonism exists between T and P, P can inhibit apoptosis. Conclusion: The metabolic balance between estrogen and progesterone and androgens is crucial for maintaining the normal function of cells. If the imbalance between local tissue proliferation and apoptosis leads to tumor occurrence.
