该文报道了纯化的强毒O17钩体39kd疏水外膜蛋白(OmpL39)在问号状钩体中的表达和对豚鼠的免疫力进行正规检定的研究.结果表明,OmpL39编码基因在问号状钩体中均有表达,但表达的量有所不同,强毒株明显多于弱毒株,同时,其抗原性也有较大差异.参照卫生部《中国生物制品规程》制造和检定钩体疫苗规程,对36只豚鼠进行三批随机对照试验.结果表明,OmpL39在豚既体内能产生高效价的阳性抗体,免疫力判定合格.经统计学处理:OmpL39组与生理盐水阴性对照组的免疫力有显著差异(P<0.05),OmpL39组与阳性对照死菌苗组的免疫保护户相当,无显著差异(P>0.05),OmpL39显示出极强的完全性免疫保护作用.本研究表明,赖型钩体O17株外膜免疫保护性抗原的分离,纯化有了新的进展,研究OmpL39编码基因和高效表达OmpL39研制新型口服OmpL39靶向控释基因疫苗已是较现实的目标.“,”In this paper,we report the expression and Identification of ImmunoprotectIon from OmpL39of L.interrogan.The Ompl39 appeared in all L.interrogans.The expressIon of OmpL39 was greater amounts in the strong virulence strains than in the weakVirutence strains.The structural gene of OmpL39 was change able in the different Servars of L.interrogans.In a ransomized control trial 36 guinea pigs were divided into OmpL39 group、Whole leptospiral cell vaccine (WLCV)group、other proteins of leptospira group and Negative coutrol group.The guinea pigs wer immunized intraperitoneally with purificated OmpL39,other proteins,WLCV and normal saline(NS).Then they were chalLenged by strain O17.The results showed that all the guinea pigs with infected OmpL39 and WLCV still survived,but all the coutrolled guinea pigs deasd.Immun Oprotective efficacy was 100% for OmpL39 and SLCV.OmpL39levels were similer as ALCV and higher than control group(P<0.05.In ten days after the second immunizingThe MAT levels of OmpL39 group wer significantly higher (1:3200)than those of WLCV group (1:1600),other proteins(1:800)and coutrol group(-).The results suggested that OmpL39could be used as important immunoprotective antigen To develop the genetic vaccine of the targeting delivery system.
