目的:探讨接骨膏外敷给药对家兔骨折愈合过程中BMPR-Ⅱ、SMAD8 m RNA表达的影响,分析其促进骨折愈合的作用机制。方法:建立30只家兔桡骨骨折模型,随机分为实验组和对照组,每组15只,实验组在其患处外敷接骨膏并用小夹板固定;对照组不使用药物,仅常规消毒后进行小夹板固定。分别于给药后第3、6、10周每组随机抽取5只,处死家兔取患处组织标本,采用实时荧光定量PCR方法测定家兔骨组织中BMPR-Ⅱ、SMAD8m RNA的含量,对所得数据进行统计学分析。结果:实验组家兔第3、10周末患处骨组织标本中BMPR-Ⅱ、SMAD8m RNA表达均高于对照组(P<0.05)。结论:接骨膏可在骨折愈合早、晚期促进患处骨组织中BMPR-Ⅱ、Smad8m RNA的表达,加速骨折愈合。
Objective: To investigate the effect of external application of Jieu Gao on expression of BMPR-Ⅱ and SMAD8 mRNA during fracture healing in rabbits, and to explore its mechanism of promoting fracture healing. Methods: Thirty rabbits with radius fracture were randomly divided into experimental group and control group, with 15 rats in each group. The experimental group was given plaster cream on the affected area and fixed with a small splint. In the control group, no drug was used, Plywood fixed. Five rabbits were randomly selected from each group at 3, 6, and 10 weeks after the administration, rabbits were sacrificed and the contents of BMPR-Ⅱ and SMAD8m RNA in rabbit bones were measured by real-time fluorescence quantitative PCR. Data for statistical analysis. Results: The expression of BMPR-Ⅱ and SMAD8m RNA in the bone tissue of the experimental group at 3 and 10 weeks were higher than that of the control group (P <0.05). Conclusion: Jiekuao can promote the expression of BMPR-Ⅱ and Smad8mRNA in the bone tissue of the affected area early and later, and accelerate the healing of fractures.
