目的:用糖皮质激素(GC)制造大鼠“阳虚”模型,以探讨其体内代谢变化。方法:16只3月龄雄性SD大鼠,体重345~347g,随机分为对照组和激素组,后者喂醋酸泼尼松4.5mg/kg,2次/周。处死大鼠前第10天和第2天,分别给其皮下注射四环素(25mg/kg)和Calcein(5mg/kg),以标明分两次注射标记物期间骨形成的情况。3个月后,用2%戊巴比妥钠(40~50mg/kg)腹腔麻醉并用注射器穿右心室抽血处死,血液送附院血液病研究室进行检测;取下的内脏精心剥离干净,湿组织称重;取胫骨上段用甲基丙烯酸甲酯行不脱钙骨包埋,制成20μ的薄片。血清碱性磷酸酶(AKP)、Ca、P、K、Cl按临床生化学检验法检测:骨片检测为半自动图象数字化分析仪,对胫骨上段松质骨的骨组织作静态和动态测量及计算。结果:与对照组比较,激素组体重减少了8.83%,内脏重量减轻,其中肾上腺和睾丸分别减少了12.12%(P<0.01)和13.66%(P<0.05);血清生化指标AKP降低了27.63%(P<0.05);胫骨上段骨小梁面积及数量分别减少38%(P<0.05)和37%(P<0.05),从而出现骨量丢失。结论:本文初步认为阳虚时?
OBJECTIVE: To establish the rat model of “yang deficiency” with glucocorticoid (GC) to explore its metabolic changes in vivo. Methods: Sixteen 3-month-old male Sprague-Dawley rats weighing 345-3 347 g were randomly divided into control group and hormone group, and the latter was fed with 4.5 mg / kg prednisone acetate twice daily. Ten days and two days prior to sacrifice, tetracycline (25 mg / kg) and Calcein (5 mg / kg) were each injected subcutaneously to indicate bone formation during two injections of the marker.
