作者采用自行设计的方法,首次在鼻咽癌病人血清中检测出凝集因子(AF)。鼻咽癌病人血清以D-Hanks缓冲液倍比稀释(1:20~2560)。人鼻咽癌细胞株(CNE-2)经胰蛋白酶消化,戊二醛固定制成单个细胞悬液。两者混合于96孔培养板,5000细胞/孔,经37℃、4℃、再37℃处理后,在倒置显微镜下观察,以能形成一个≥20细胞的凝集团块为AF阳性,结果发现231例鼻咽癌病人血清都是阳性,几何平均滴度(GMT)1:265.1。111名正常人中74人AF阴性,37人低滴度阳性(1:20~160),GMTI:39.2。提示鼻咽癌病人血清AF水平明显高于正常人(P<0.001)。作者对结果及方法进行了讨论。
The authors used a self-designed method for the first time detected in the serum of patients with nasopharyngeal carcinoma agglutination factor (AF). Nasopharyngeal carcinoma patients were serially diluted in D-Hanks buffer (1: 20-2560). Human nasopharyngeal carcinoma cell line (CNE-2) was trypsinized and fixed with glutaraldehyde to make a single cell suspension. The two were mixed in 96-well culture plates, 5000 cells / well, after 37 ℃, 4 ℃, then 37 ℃ under the inverted microscope to observe, in order to form a ≥ 20 cell agglutination mass was AF positive, found that 231 cases of nasopharyngeal carcinoma patients were all seropositive, the geometric mean titer (GMT) 1: 265.1.111 74 normal AF negative, 37 low-titer positive (1:20 ~ 160), GMTI: 39.2.
