目的:优化枸杞甜菜碱提取物的液相色谱分析条件。方法:采用反相高效液相色谱法对枸杞甜菜碱的检测波长、流动相进行了优化。结果色谱柱为江苏汉邦lichrospher NH 2 250mm×4.6mm(5μm),流动相为乙腈:水(85:15),等度洗脱,流速:0.7 m L·min-1,检测波长:195nm,柱温:30℃,进样量5μL。精密度、稳定性、重现性实验色谱峰相对保留时间分别为RSD%﹤0.9%、RSD%﹤0.79%、RSD%﹤0.84%;相对峰面积均RSD%﹤3%。结论:采用乙腈体系为枸杞甜菜碱色谱分析的流动相,等度洗脱,甜菜碱保留时间适中,分离效果好,方法精密度、稳定性、重现性良好。可为高效液相色谱法检测枸杞甜菜碱提供一定参考。
Objective: To optimize the liquid chromatography conditions of Lycium barbarum extract. Methods: The detection wavelength and mobile phase of Lycium barbarum were optimized by RP-HPLC. Results The chromatographic column was characterized by lichrospher NH 2 250 mm × 4.6 mm (5 μm) with a mobile phase of acetonitrile: water (85:15), isocratic elution at a flow rate of 0.7 mL · min-1 and the detection wavelength was 195 nm. Column temperature: 30 ℃, injection volume 5μL. The RSD% <0.9%, RSD% <0.79%, RSD% <0.84%, relative peak area RSD% <3%, respectively. CONCLUSION: Acetonitrile is the mobile phase of Lycium barbarum chromatographic analysis with isocratic elution. The retention time of betaine is moderate, and the separation effect is good. The precision, stability and reproducibility of the method are good. It can provide reference for the determination of Lycium betaine by high performance liquid chromatography.
