Topical transdermal formulations developed for methotrexate (MTX) are good for topical treatment of psoriasis. Therefore, the effects of different solvent dilution and prescription factors on the conversion of MTX precursor vesicle gels to vesicles were investigated. A series of MTX precursor vesicle gels consisting of Span-40, Chol and Tween-20 (T20) were designed and prepared by Box-Behnken. The vesicle size, MTX entrapment efficiency and zeta potential were evaluated Indicators to examine the dilution effect of short chain alcohols (ie ethanol, propylene glycol and glycerol) on precursor vesicles. The results show that the MTX precursor vesicle gel hydrated in the test solvent
