研究响应面法与酶法联用工艺提取拐枣中的活性物质拐枣多糖及其抗氧化活性。采用辅助热水提取法提取拐枣多糖,以Box-Behnken试验设计结合响应面分析法优化了提取工艺条件,确定最佳工艺参数。结果显示,在酶解温度为50℃,酶解浓度为0.06 g和p H为5.0的条件下,拐枣多糖的最高含量为19.79%。拐枣多糖具有明显的抗氧化作用和清除自由基的作用,当质量浓度为5.0 mg·m L-1时,FRAP值为1.91,对2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐自由基、1,1-二苯基-2-苦基肼基自由基、羟基自由基和超氧阴离子自由基的清除率分别达到69.63%、74.10%、44.13%和73.79%。
Study the response surface method and enzymatic combined process of Chinese jujube extract active substances jujube polysaccharide and its antioxidant activity. Auxiliary hot water extraction method was used to extract jujube polysaccharide. The Box-Behnken experimental design combined with response surface methodology was used to optimize the extraction process conditions and determine the optimum process parameters. The results showed that the highest content of jujube polysaccharides was 19.79% under the conditions of enzymolysis temperature of 50 ℃, enzymolysis concentration of 0.06 g and p H of 5.0. Curcuma longa polysaccharide has obvious anti-oxidation and scavenging free radicals. When the concentration of 5.0 mg · m L-1, the FRAP value was 1.91, and 2,2-azino-bis (3-ethyl- Thiazole-6-sulfonic acid) diammonium salt radical, 1,1-diphenyl-2-picrylhydrazyl radical, hydroxyl radicals and superoxide anion radical scavenging rate reached 69.63%, 74.10% , 44.13% and 73.79% respectively.
