为验证抑癌基因RB的抑癌作用及进一步探讨其抑癌机理,我们用带有人RB基因cDNA的真核细胞表达质粒转染小鼠乳腺上皮恶性转化细胞系11A1,得到4个稳定的逆转细胞系11A1-R1~R4。逆转细胞在形态、核质比例上接近正常细胞,在琼脂表面生长能力下降,裸鼠体内致瘤力下降。RT-PCR及Northern杂交结果显示逆转细胞中RB基因表达增高,c-myc基因表达水平下降。本实验证明了RB基因的抑癌作用并建立了一个良好的上皮型细胞表型逆转的模型。
In order to verify the tumor suppressing effect of the tumor suppressor gene RB and further explore its tumor suppressing mechanism, we transfected the mouse mammary epithelial malignant transformed cell line 11A1 with the human eukaryotic expression plasmid containing the human RB gene cDNA to obtain four stable reversing cells. Department 11A1-R1~R4. Reversing the cell morphology and nucleus-to-cytoplasm ratio was similar to that of normal cells. The ability to grow on the surface of agar decreased, and tumorigenicity in nude mice decreased. The results of RT-PCR and Northern hybridization showed that the expression of RB gene was increased in the reversing cells, and the expression level of c-myc gene was decreased. This experiment demonstrated the tumor suppressing effect of RB gene and established a good model of epithelial cell phenotype reversal.
