目的 探讨中动脉梗塞大鼠脑缺血 再灌注后IL 1 β、c fos、促肾上腺皮质激素释放因子CRF在脑内海马区表达的时间规律及其时序关系。方法 采用大脑中动脉插入丝线结扎 (MCAO)方法制造大鼠脑缺血 再灌注缺血模型。分别按照 30min至 7d的不同灌注时间取材 ,假手术组大鼠作为对照组。应用免疫组化方法标记实验各组大鼠脑组织中海马区IL 1 β、c fos、CRF阳性神经元数量。 结果 免疫组化结果表明 ,IL 1 β免疫活性细胞在缺血再灌注后从 1h(97 4± 39 3)后开始表达 ,2h(61 0 67± 1 88 4)后达高峰 ,至 7d(6 6± 5 9)时IL 1 β免疫活性细胞表达基本消失 ;c fos免疫活性细胞从 2h(1 62 4± 2 4 75)开始表达 ,4h(52 1 7± 1 0 3 5)达高峰 ,4d(1 0 4± 9 7)时表达基本消失 ;CRF免疫活性细胞从 2h时 (97 2± 1 8 4)开始表达 ,1 2h(374 4± 56 95)和 7d(396 3± 50 98)时表达分别达到 2次高峰。脑缺血 再灌注后IL 1 β、c fos、CRF在海马区表达高峰时间具有一定的前后时序 ,即IL 1 β表达早于c fos,而CRF表达随c fos之后。 结论 大鼠脑缺血 再灌注后可诱发IL 1 β、c fos、CRF表达增加 ,具有一定时间规律性和时序关系。
Objective To investigate the temporal regularity of IL-1β, c fos and corticotropin-releasing factor (CRF) expression in the hippocampus of rats with middle cerebral artery occlusion (MCAO) and their time-series relationship. Methods Cerebral ischemia - reperfusion ischemia was induced by middle cerebral artery occlusion (MCAO) in rats. According to the different perfusion time from 30min to 7d, the rats in the sham operation group were taken as the control group. The number of IL-1β, c fos and CRF positive neurons in the hippocampus in each group were marked by immunohistochemistry. Results Immunohistochemical results showed that IL 1 β immunocompetent cells began to express after 1 h (97 4 ± 39 3) and peaked at 2 h (61 0 67 ± 1 88 4), and reached peak at 7 d 6 ± 5 9), the expression of IL 1 β immunocompetent cells disappeared. The expression of c fos immunoreactive cells began to express at 2 h (1 62 4 ± 2 4 75), peaked at 4 h (52 1 7 ± 1 0 3 5) (1 0 4 ± 9 7), the expression of CRF immunoreactive cells began to expire at 2 h (97 2 ± 1 8 4), while the expression of CRF immunoreactive cells at 2 h (374 4 ± 56 95) and 7 d (396 3 ± 50 98) The expression reached the second peak respectively. After cerebral ischemia-reperfusion, the expression of IL-1β, c fos and CRF in hippocampus had a certain time-sequence, that is, the expression of IL-1β was earlier than that of c fos and the expression of CRF was following c fos. Conclusion The expression of IL-1β, c-fos and CRF may be induced after cerebral ischemia-reperfusion in rats, with a certain regularity of time and the relationship of timing.
