目的:探讨荷瘤小鼠体内硒水平,谷胱甘肽过氧化物酶(GSH-Px)活力及丙二醛(MDA)含量的变化和亚硒酸钠对这种变化的影响。方法:采用DAN荧光法、DTNB比色法及TBA荧光法分别测定荷瘤及正常小鼠体内微量元素硒的含量、GSH-Px活力及过氧化脂质产物MDA的含量。结果:发现荷瘤小鼠体内全血及肝组织中硒的含量、肝匀浆中GSH-px活力均明显低于正常小鼠。亚硒酸钠虽然在该实验剂量下对S-180肉瘤的生长没有显著的抑制作用,但能纠正荷瘤小鼠全血及肝组织中硒含量的下降,增加全血及肝组织中GSH-Px活力,降低血浆中MDA的含量,对抗CCl4引起的肝组织中GSH-px活力下降。结论:亚硒酸钠能对抗肿瘤对机体抗氧化能力的削弱作用,这时肿瘤的预防和治疗有一定意义。
Objective: To investigate the changes of selenium level, glutathione peroxidase (GSH-Px) activity and malondialdehyde (MDA) content in tumor-bearing mice and the effect of sodium selenite on the changes. Methods: The content of trace element selenium, the activity of GSH-Px and the content of lipid peroxidation product MDA in tumor-bearing and normal mice were detected by DAN fluorescence method, DTNB colorimetry and TBA fluorescence method respectively. Results: The content of selenium and the activity of GSH-px in liver homogenate of whole blood and liver tissue of tumor-bearing mice were significantly lower than that of normal mice. Although sodium selenite had no significant inhibitory effect on the growth of S-180 sarcoma at the experimental dose, it could correct the decrease of selenium content in whole blood and liver tissue of tumor-bearing mice and increase the content of GSH- Px activity, reduce the content of MDA in the plasma, against CCl4-induced liver tissue GSH-px activity decreased. Conclusion: Sodium selenite can counteract the anti-tumor effect on the antioxidant capacity of the body, then the prevention and treatment of cancer have some significance.
