EB病毒(EBV)转化的B细胞具有优势分泌人IgH的特点。本文应用Panning法分离正常人外周血中SmIgM~+和SmIgG~+B细胞亚群,分别用EBV转化,再与人-鼠骨髓瘤细胞系SHM-D33融合,以转化率和融合率两个指标比较两种B细胞亚群在转化和融合水平的差异。结果表明,SmIgM~+B细胞亚群对EBV转化的敏感性是SmIgG~+B细胞亚群的4~5倍:而融合率无显著差异。部分解释了EBV转化和EBV-杂交瘤技术中人IgM优势分泌现象的原因。所采用的Panning技术还可用于定向生产IgG类人单抗。
Epstein-Barr virus (EBV) -transformed B cells have the predominant feature of secreting human IgH. In this paper, the Panning method was used to separate the SmIgM ~ + and SmIgG ~ + B cell subsets in normal human peripheral blood, which were respectively transformed into EBV and then fused with human myeloma cell line SHM-D33. The transformation rate and fusion rate Differences in the levels of transformation and fusion between the two B cell subsets were compared. The results showed that the sensitivity of SmIgM ~ + B cell subpopulation to EBV transformation was 4 ~ 5 times higher than that of SmIgG ~ + B cell subpopulation: the fusion rate had no significant difference. In part explains the reasons for the superior secretion of human IgM in EBV transformation and EBV-hybridoma technology. The Panning technology used can also be used to target the production of IgG human monoclonal antibodies.
