survivin,凋亡蛋白抑制因子(IAP)家族中的重要成员之一,对细胞的有丝分裂和细胞程序性死亡有重要的调节作用。起初,survivin被描述为caspase-9的抑制因子,随着近些年的研究发现survivin的功能不只限于肿瘤的凋亡抑制,还有对有丝分裂关键点、促血管生成和化疗耐药有调控作用。野生型p53抑制survivin基因的转录表达,而肿瘤中存在很多机制能解除对survivin的抑制,如基因扩增、甲基化、启动子活性增强和p53的功能丧失。现对survivin在细胞凋亡和肿瘤发生中的作用以及新型抗肿瘤因子-survivin抑制因子的研究进展做一综述。
Survivin, one of the important members of the inhibitor of apoptosis protein (IAP) family, plays an important regulatory role in cell mitosis and apoptosis. Initially, survivin was described as an inhibitor of caspase-9. In recent years, survivin has been found to function not only in the inhibition of tumor apoptosis, but also in the regulation of mitosis, angiogenesis and chemoresistance. Wild-type p53 inhibits the transcriptional expression of survivin, and there are many mechanisms in the tumor that can relieve the inhibition of survivin, such as gene amplification, methylation, promoter activity enhancement and p53 loss of function. The role of survivin in apoptosis and tumorigenesis as well as the progress of the new anti-tumor factor-survivin inhibitor are reviewed.
