目的依据孕妇血浆中存在游离胎儿DNA的理论,从孕妇外周血浆中分离出胎儿DNA并加以鉴定,探讨前置胎盘孕妇外周血中胎儿DNA含量的变化及其临床意义。方法采用实时荧光定量PCR(real-time fluorescence quantitative Polymerase chain-reaction,FQ-PCR)方法对35例明确为前置胎盘孕妇血浆中胎儿DNA的含量进行检测并比较,初步探讨游离f DNA在不同状态下的浓度变化。结果前置胎盘粘连组、前置胎盘非粘连组及对照组孕妇血浆中DNA的含量拷贝数分别为(630.34±75.52)、(490.08±56.32)、(230.82±25.47)copys/m I。各组间孕妇外周血中胎儿DNA的含量比较,差异有统计学意义(P<0.05)。结论孕妇外周血中胎儿DNA量与胎盘组织的功能状态有关。测定孕妇外周血中胎儿DNA量可能有助于预测前置胎盘合并胎盘粘连。
OBJECTIVE According to the theory of free fetal DNA in pregnant women’s plasma, fetal DNA was isolated from peripheral blood of pregnant women and identified, to investigate the changes of fetal DNA content in peripheral placental blood and its clinical significance. Methods FQ-PCR was used to detect and compare the content of fetal DNA in plasma of 35 pregnant women with placenta previa, and to explore the relationship between the content of free f DNA in different states Under the concentration changes. Results The copy number of DNA in the placenta adhesion group, placenta previa non-adhesion group and control group were (630.34 ± 75.52), (490.08 ± 56.32) and (230.82 ± 25.47) copys / m I. The fetal DNA content of pregnant women in each group was significantly different (P <0.05). Conclusion The amount of fetal DNA in peripheral blood of pregnant women is related to the functional status of placenta. Determining the amount of fetal DNA in a pregnant woman’s peripheral blood may be helpful in predicting placenta previa placenta adhesion.
