补肾化痰活血中药对再生障碍性贫血小鼠骨髓CD_(34)~+和细胞因子表达的影响

目的:观察和比较补肾化痰活血中药对再生障碍性贫血小鼠骨髓造血生长因子SCF mRNA、GM-CSF mRNA表达和骨髓CD34+的影响。方法:建立再障小鼠模型,各组小鼠分别胃饲生理盐水、补肾化痰活血中药、康力龙和再障生血片,连续喂养14天后,采集骨髓进行单个核细胞培养,应用流式细胞术检测骨髓CD3+4抗原水平;应用RT-PCR方法检测SCFmRNA、GM-CSF mRNA表达水平。结果:①模型组骨髓CD3+4抗原检测结果显著低于正常对照组,治疗后各组小鼠骨髓CD3+4抗原水平明显提高,差异有显著性。②模型组骨髓造血生长因子SCF mRNA、GM-CSF mRNA表达水平均低于正常对照组,治疗后各组小鼠骨髓造血生长因子表达水平均明显提高,差异均有显著性。结论:补肾化痰活血方可能通过增强再障小鼠骨髓造血生长因子GM-CSFmRNA、SCFmRNA表达水平,刺激造血细胞分化成熟,达到调控骨髓造血的目的。 Objective: To observe and compare the effects of Bushen Huayu Huoxue Chinese Medicine on bone marrow hematopoietic growth factor SCF mRNA, GM-CSF mRNA expression and bone marrow CD34+ in mice with aplastic anemia. METHODS: A mouse model of aplastic anemia was established. Each group of mice was administrated with saline, supplemented with kidney and phlegm and blood circulation herbs, stanozolone, and Zaozhuang Shengxue tablets. After 14 consecutive days of feeding, bone marrow was collected for mononuclear cell culture and flow cytometry was used. The level of CD3+4 antigen in bone marrow was measured; the expression of SCFmRNA and GM-CSF mRNA was detected by RT-PCR. RESULTS: 1The detection results of bone marrow CD3+4 antigen in the model group were significantly lower than that in the normal control group. After treatment, the CD3+4 antigen level in the bone marrow of the mice in each group was significantly increased, and the difference was significant. 2 The expression of bone marrow hematopoietic growth factor (SCF) mRNA and GM-CSF mRNA in the model group was lower than that in the normal control group. The expression of bone marrow hematopoietic growth factors in each group was significantly increased after treatment, and the differences were significant. Conclusion: Bushen Huayu Huoxue Recipe may enhance the expression of bone marrow hematopoietic growth factor GM-CSF mRNA and SCF mRNA in aplastic anemia mice, and stimulate the differentiation and maturation of hematopoietic cells to achieve the purpose of regulating bone marrow hematopoiesis.
