目的:探讨雷公藤内酯醇(TP)对试验性色素膜视网膜炎(EAU)Th1反应相关因子的抑制作用。方法:用ELISA检测IL-12、IFN-γ水平;采用原位杂交染色法,观测TP对EAU的T淋巴细胞IL-12mRNA的表达的影响;采用电泳迁移率改变试验对EAU T淋巴细胞NF-AT活性检测。结果:①TP能有效的降低S-抗原诱导产生的EAU(P<0.05);②TP能降低EAU T淋巴细胞IL-12、IFN-γ分泌水平;③P能抑制EAT淋巴细胞IL-12 mRNA的表达;④TP抑制EAU T淋巴细胞NF-AT活性。结论:TP对EAU的免疫抑制作用可能是通过抑制与Th1反应相关的调节因子使Th1反应减弱,而使EAU的发病率明显降低。
Objective: To investigate the inhibitory effect of triptolide (TP) on Th1 response-related factors in experimental retinitis pigmentosa (EAU). Methods: The levels of IL-12 and IFN-γ were detected by ELISA. The effect of TP on the expression of IL-12 mRNA in T lymphocytes of EAU was observed by in situ hybridization. The electrophoretic mobility shift assay AT activity test. Results: ①TP could effectively reduce the EAU induced by S-antigen (P <0.05); ②TP could decrease the secretion of IL-12 and IFN-γ in EAU T lymphocytes; ③P could inhibit the expression of IL-12 mRNA in EAT lymphocytes; ④ TP inhibit EAU T lymphocyte NF-AT activity. Conclusion: The immunosuppressive effect of TP on EAU may be due to the inhibition of Th1 response-related regulatory factors to reduce the Th1 response, leaving the incidence of EAU significantly reduced.
