中药半夏(Pinellia ternate(Thunb.)Breit)是常用的中药,使用广泛,安徽是半夏的道地产区之一,但由于过度采集,其野生资源日渐匮乏,为了保持野生资源的持续性发展,物种的保护刻不容缓.我们的研究就是以安徽半夏“三珍”之一的颖半夏作为研究对象,探索颖半夏种质胚性愈伤的离体保存方法,以颖半夏胚性愈伤为材料,考察植物生长调节剂、温度、大量元素及蔗糖对胚性愈伤保存时间、保存材料再生苗形态学、生理生化指标的影响.在培养室温度10℃、光照度1 500 Lx、每日光照时间12 h的环境下,颖半夏胚性愈伤在培养基 1/2 MS+Sucrose 40.0 g/L+CCC 0.1 g/L+ABA 1.0 mg/L+PP333 2.0 mg/L 中保存 9 个月,存活率超过 70%,保存材料再生苗形态学、部分生理生化指标与正常继代苗无明显差异.本研究实现了道地中药颖半夏种质胚性愈伤的中期离体保存,为颖半夏种质资源保存提供了有效途径.“,”Pinellia tub er is a common traditional Chinese medicine widely used in clinic. One of the genuine producing areas of Pinellia tuber is in Anhui. Due to excessive collection, its wild resources are increasingly scarce, and the protection of the species is urgently needed in order to keep the wild resources sustainable development. Our research was to explore a method for germplasm embryogenic callus preservation in vitro of Pinellia tuber. The embryogenic calluses of Pinellia tuber were used as experimental materials to study the effects of plant growth regulator, temperature, large number of elements in MS medium and sucrose on the preservation time of embryogenic calluses, and morphology and some physiological and biochemical indexes of regenerated plantlet from preservation material. When embryogenic calluses of Pinellia tuber were inoculated onto 1/2 MS media containing 40.0 g/L sucrose, and supplemented with 0.1 g/L CCC, 1.0 mg/L ABA, and 2.0 mg/L PP333 in an illuminated chamber under a 12 h photoperiod of 1 500 Lx light intensity at 10℃ for nine months, the survival rate of embryogenic calluses was over 70%, and morphology, and some physiological and biochemical indexes of regenerated plantlet from preservation material were not significantly different from subculture plantlet. The study achieved medium-term germplasm embryogenic callus preservation in vitro of Pinellia tuber, which might provide an effective way for germplasm preservation of Pinellia tuber.
