HL_(60)细胞经二甲基亚砜(DMSO)作用6天后,85%的细胞发生成熟样改变:胞体变小,胞浆颗粒增多,核凹陷,核仁模糊不清;溶菌酶分泌增加,克隆率降低;吞噬功能增强。但相当部分成熟样细胞仍有核仁,去除DMSO,进一步培养时,又恢复原来形及及增殖能力。由此认为,DMSO对HL_(60)细胞的诱导分化作用是不完全的,且具有可逆性。为从根本上改变此细胞的肿瘤特性,需探索更有效的诱导剂与诱导方法。
Eighty-five percent of cells in HL_ (60) cells treated with dimethylsulfoxide (DMSO) for 6 days showed mature changes: the cytoplasm became smaller, cytoplasmic granules increased, the nuclei became hollow and the nucleolus became vague; the secretion of lysozyme increased, Clone rate decreased; phagocytosis enhanced. However, a considerable portion of mature cells still have nucleolus, remove the DMSO, further culture, and restore the original shape and proliferation. Thus, DMSO induced differentiation of HL 60 cells is incomplete and reversible. In order to fundamentally change the tumor characteristics of this cell, it is necessary to explore more effective inducer and induction method.
