目的:从尿液浓缩机制方面研究六味地黄汤的滋阴作用机制。方法:36只成年雄性SD大鼠分为正常对照组、模型组和中药组。采用放免法测定血浆血管升压素(AVP)水平、免疫组化法检测肾髓质水通道蛋白2(AQP2)表达。结果:与正常对照组相比,模型组大鼠的体重、尿量明显减少,AVP水平上升,AQP2表达增加;中药组上述指标恢复到接近正常组或正常组水平。结论:肾阴虚时机体尿液浓缩机制亢进,六味地黄汤的滋阴作用与其抑制亢进的尿液浓缩机制有关。
Objective: To study the mechanism of nourishing Yin from Liuwei Dihuang Decoction from the aspect of urine concentration mechanism. Methods: Thirty-six adult male Sprague Dawley rats were divided into normal control group, model group and TCM group. The level of plasma vasopressin (AVP) was measured by radioimmunoassay and the expression of aquaporin 2 (AQP2) in renal medulla was detected by immunohistochemistry. Results: Compared with the normal control group, the body weight and urine volume of the model group decreased significantly, while the level of AVP increased and the expression of AQP2 increased. In the traditional Chinese medicine group, the above indexes returned to normal levels or normal levels. CONCLUSION: The mechanism of urine concentration in the body of the kidney and yin deficiency is related to the mechanism of urine concentration of Liuweidihuang Decoction and its inhibition of hyperthyroidism.
