目的探讨孕期铅水平对胎盘组织NF-κb表达和细胞凋亡的影响,了解铅的胚胎毒性机制。方法 67例孕妇于分娩时抽取外周血,同时抽取脐血并收集胎盘组织。原子吸收光谱法测定孕期血铅水平;免疫组织化学方法测定NF-κb在胎盘中的表达。Hoechst法和TUNEL法检测胎盘细胞凋亡指数。结果血铅水平与胎盘组织中NF-κb表达和细胞凋亡指数显著相关,相关系数分别为:0.663和0.614,P<0.01。结论铅暴露引起NF-κb表达异常可能是胎盘细胞凋亡失衡和胚胎发育异常的重要机理之一。
Objective To investigate the effect of lead levels during pregnancy on the expression of NF-κB and apoptosis in placentas, and to understand the embryo toxicity mechanism of lead. Methods 67 cases of pregnant women during childbirth to extract peripheral blood, while taking cord blood and placental tissue collection. Blood lead level was measured by atomic absorption spectrometry during pregnancy. The expression of NF-κB in the placenta was determined by immunohistochemistry. Hoechst method and TUNEL method were used to detect the apoptosis index of placenta. Results There was a significant correlation between blood lead levels and the expression of NF-κB and apoptosis in placentas. The correlation coefficients were 0.663 and 0.614, respectively, P <0.01. Conclusion The abnormal expression of NF-κB induced by lead exposure may be one of the important mechanisms of imbalanced placental apoptosis and embryonic development.
