目的:探讨唑来膦酸联合白细胞介素-2(IL-2)诱导胃癌患者外周血γδT细胞的作用。方法:收集胃癌患者和健康捐献者各30例,分别采集外周静脉血并分离出单核细胞(PBMC),采用流式细胞仪(FCM)检测唑来膦酸联合IL-2诱导前后γδT细胞的数量及其CD69分子的表达情况。结果:胃癌组的γδT细胞数量和百分比均明显低于健康对照组(P<0.05);唑来膦酸联合IL-2诱导PBMC后,胃癌实验组和健康对照组中的γδT细胞的数量和百分比均较诱导前明显升高(P<0.05)。结论:唑来膦酸联合IL-2体外诱导PBMC可显著扩增γδT细胞,以加强杀伤肿瘤细胞的目的。
Objective: To investigate the effect of zoledronic acid combined with interleukin-2 (IL-2) on the induction of γδT cells in peripheral blood of patients with gastric cancer. Methods: Thirty patients with gastric cancer and healthy donors were collected. Peripheral venous blood was collected and mononuclear cells (PBMCs) were collected. Flow cytometry (FCM) was used to detect the expression of γδT cells before and after the induction of IL-2 Quantity and expression of its CD69 molecule. Results: The number and percentage of γδT cells in gastric cancer group were significantly lower than those in healthy control group (P <0.05). After the PBMCs were induced by zoledronic acid and IL-2, the number and percentage of γδT cells in gastric cancer group and healthy control group Were significantly higher than before induction (P <0.05). Conclusion: Zoledronic acid combined with IL-2 can induce γδT cells to proliferate significantly in vitro, so as to enhance the killing effect on tumor cells.
