目的:检测高迁移率族蛋白A(HMGA)在雄性小鼠生殖细胞株TM4、GC-1spg、GC-2spd(ts)中的表达,为深入探讨HMGA基因在小鼠精子发生过程中的作用机制奠定理论基础。方法:应用RT-PCR、Western印迹检测雄性小鼠生殖细胞株TM4、GC-1spg、GC-2spd(ts)中HMGA1、HMGA2基因的表达。结果:HMGA1mRNA、HMGA2mRNA在雄性小鼠生殖细胞株TM4、GC-1spg、GC-2spd(ts)中均有不同程度表达,Western印迹结果表明HMGA1与HMGA2蛋白在雄性小鼠生殖细胞株TM4、GC-1spg、GC-2spd(ts)中呈阳性表达,与RT-PCR的结果一致。结论:HMGA的表达可能参与了细胞株TM4、GC-1spg、GC-2spd(ts)的分裂、增殖,在雄性小鼠精子发生过程中可能发挥着重要作用。
OBJECTIVE: To detect the expression of high mobility group box protein (HMGA) in TM4, GC-1spg and GC-2spd (ts) of male mouse germ cell lines. To explore the mechanism of HMGA gene in mouse spermatogenesis Lay the theoretical foundation. Methods: The expression of HMGA1 and HMGA2 genes in TM4, GC-1spg and GC-2spd (ts) of male mice germ cell line were detected by RT-PCR and Western blot. Results: HMGA1mRNA and HMGA2mRNA were expressed in different degrees in TM4, GC-1spg and GC-2spd (ts) of germ cell lines of male mice. Western blot results showed that HMGA1 and HMGA2 protein were expressed in the male mouse germ cell lines TM4, GC- 1spg, GC-2spd (ts) positive expression, consistent with the results of RT-PCR. CONCLUSION: The expression of HMGA may be involved in the division and proliferation of cell lines TM4, GC-1spg and GC-2spd (ts) and may play an important role in the spermatogenesis of male mice.
