[目的]研究STR基因座DYS452在延边地区朝鲜族和汉族人群中的遗传多态性.[方法]设计DYS452荧光标记引物,利用PCR扩增与ABI310基因分析仪对无关联的各200份朝鲜族和汉族个体的DYS452基因座进行检测,用GeneMapperID v3.2软件分析电泳结果并分析获得群体遗传多态性数据.[结果]朝鲜族和汉族人群在DYS452基因座上共检测出7个等位基因,朝鲜族人群中检测出5个等位基因,分别为29~33号等位基因;汉族人群中检测出7个等位基因,分别为27~33号等位基因.朝鲜族和汉族人群的GD值分别是0.660 7和0.696 4.将DYS452基因座在朝鲜族和汉族中的遗传分布分别与不同地区、民族及种族进行比较,获得不同群体间的P值和遗传距离.[结论]DYS452基因座在延边地区朝鲜族和汉族群体中具有较高的遗传多态性,可为该地区的法医学个人识别、亲权鉴定及人类遗传学调查提供依据.
[Objective] To investigate the genetic polymorphism of STR loci DYS452 in Korean and Han nationality in Yanbian area. [Methods] DYS452 fluorescently labeled primers were designed and used to detect the polymorphism of DYS452 in unrelated 200 Korean ethnicity And DYS452 locus in Han individuals were detected by GeneMapperID v3.2 software analysis of electrophoresis results and analysis of population genetic polymorphism data. [Results] Korean and Han populations in the DYS452 locus detected a total of seven alleles , 5 alleles were detected in Korean population, which were 29-33 alleles respectively; 7 alleles were detected in Han population, which were 27-33 alleles respectively. GD values were 0.660 7 and 0.696, respectively.4 The genetic distributions of DYS452 loci in Korean and Han were compared with those in different regions, ethnic groups and races separately to obtain the P value and genetic distance among different groups. [Conclusion] The DYS452 gene Block in Yanbian Korean ethnic group and Han population has a high genetic polymorphism, forensic science in the region for personal identification, paternity testing and human genetics provide the basis for investigation.
